DESCRIPTION: RecName: Full=Golgin subfamily A member 5; AltName: Full=Cell proliferation-inducing gene 31 protein; AltName: Full=Golgin-84; AltName: Full=Protein Ret-II; AltName: Full=RET-fused gene 5 protein;
FUNCTION: Involved in maintaining Golgi structure. Stimulates the formation of Golgi stacks and ribbons. Involved in intra-Golgi retrograde transport.
SUBUNIT: Homodimer. Interacts with RAB1A that has been activated by GTP-binding, and possibly also with OCRL1. Interacts with isoform CASP of CUX1.
SUBCELLULAR LOCATION: Golgi apparatus membrane; Single-pass type IV membrane protein. Note=Found throughout the Golgi, both on cisternae and, at higher abundance, on the tubulo-vesicular structures of the cis-Golgi network.
TISSUE SPECIFICITY: Ubiquitous. Highly expressed in seminiferous tubules and Leydig cells in testis, and detected at much lower levels in the other tissues tested. Expression is very low or not detectable in spermatozoa.
PTM: Highly phosphorylated during mitosis. Phosphorylation is barely detectable during interphase.
DISEASE: Defects in GOLGA5 are a cause of thyroid papillary carcinoma (TPC) [MIM:188550]. TPC is a common tumor of the thyroid that typically arises as an irregular, solid or cystic mass from otherwise normal thyroid tissue. Papillary carcinomas are malignant neoplasm characterized by the formation of numerous, irregular, finger-like projections of fibrous stroma that is covered with a surface layer of neoplastic epithelial cells. Note=A chromosomal aberration involving GOLGA5 is found in thyroid papillary carcinomas. Translocation t(10;14)(q11;q32) with RET. The translocation generates the RET/GOLGA5 (PTC5) oncogene which was found in 2 cases of PACT in children exposed to radioactive fallout after Chernobyl.
