DESCRIPTION: RecName: Full=Mitogen-activated protein kinase kinase kinase 3; EC=2.7.11.25; AltName: Full=MAPK/ERK kinase kinase 3; Short=MEK kinase 3; Short=MEKK 3;
FUNCTION: Component of a protein kinase signal transduction cascade. Mediates activation of the NF-kappa-B, AP1 and DDIT3 transcriptional regulators.
CATALYTIC ACTIVITY: ATP + a protein = ADP + a phosphoprotein.
COFACTOR: Magnesium.
ENZYME REGULATION: Activated by phosphorylation on Thr-530 (By similarity).
SUBUNIT: Binds both upstream activators and downstream substrates in multimolecular complexes. Part of a complex with MAP2K3, RAC1 and CCM2. Interacts with MAP2K5 and SPAG9.
INTERACTION: Q6Q0C0:TRAF7; NbExp=3; IntAct=EBI-307281, EBI-307556;
PTM: Phosphorylation at Ser-166 and Ser-337 by SGK1 inhibits its activity.
SIMILARITY: Belongs to the protein kinase superfamily. STE Ser/Thr protein kinase family. MAP kinase kinase kinase subfamily.
SIMILARITY: Contains 1 OPR domain.
SIMILARITY: Contains 1 protein kinase domain.

The pictures above may be empty if there is no ModBase structure for the protein. The ModBase structure frequently covers just a fragment of the protein. You may be asked to log onto ModBase the first time you click on the pictures. It is simplest after logging in to just click on the picture again to get to the specific info on that model.

-  Orthologous Genes in Other Species
  Orthologies between human, mouse, and rat are computed by taking the best BLASTP hit, and filtering out non-syntenic hits. For more distant species reciprocal-best BLASTP hits are used. Note that the absence of an ortholog in the table below may reflect incomplete annotations in the other species rather than a true absence of the orthologous gene.
