FUNCTION: May have an important and specific function in tumor progression and embryogenesis. Cleaves alpha-1-antitrypsin.
COFACTOR: Binds 1 zinc ion per subunit (By similarity).
COFACTOR: Calcium (By similarity).
SUBCELLULAR LOCATION: Secreted (Potential).
TISSUE SPECIFICITY: Identified in fetal brain, kidney and liver. In adult tissues found primarily in ovary, kidney, liver, lung, placenta, brain and peripheral blood leukocytes. Expressed as well in various cancer cell lines.
DOMAIN: The conserved cysteine present in the cysteine-switch motif binds the catalytic zinc ion, thus inhibiting the enzyme. The dissociation of the cysteine from the zinc ion upon the activation-peptide release activates the enzyme.
PTM: The precursor is cleaved by a furin endopeptidase (By similarity).
SIMILARITY: Belongs to the peptidase M10A family.
SIMILARITY: Contains 4 hemopexin-like domains.
